Megazyme™ Lactose Assay Kit (Sequential/High Sensitivity)

Enzymatic analysis for the quantitative determination of lactose in food, specifically adapted for low-lactose and lactose-free products. The method is based on a sequential enzymatic reaction in which free glucose is first removed, and lactose is then quantified via the formation of NADPH at 340 nm.

Description

The Megazyme™ Lactose Assay Kit (K-LOLAC) is designed for the specific and highly sensitive analysis of lactose in dairy products and other foods. The method is specifically optimized for low-lactose and lactose-free products, where traditional enzymatic methods often result in overestimation.

The analysis is based on a sequential enzymatic principle in which free D-glucose is first removed, followed by the hydrolysis of lactose using β-galactosidase. The released glucose is quantified via a coupled enzymatic reaction (HK/G6PDH/6PGDH) by measuring NADPH at 340 nm.

This method enables the selective detection of lactose even in the presence of galacto-oligosaccharides (GOS), which is essential for the accurate analysis of lactose-free products.

The kit has been validated in accordance with AOAC Method 2020.08 and meets the accuracy and reproducibility requirements for food analysis.

Features

  • Sequential analysis of free glucose and lactose in the same cuvette
  • High specificity for lactose (minimal interference from GOS)
  • Very low detection limit (~1.6 mg/L)
  • Optimized for low-lactose and lactose-free products
  • AOAC method (August 2020)
  • Reproducible and robust analysis
