Reagent D is a photoreactive PCR designed to remove DNA from dead cells, enabling the selective detection of live microorganisms and reducing false-positive results.
Description
Reagent D is used as a pre-analytical step in PCR and LAMP microbiological analysis to enable the selective detection of live microorganisms. The reagent contains a photoreactive substance that penetrates cells with damaged membranes (dead cells) and, after light exposure, covalently binds to DNA. This blocks amplification in subsequent PCR, eliminating signals from non-viable DNA and reducing the risk of false-positive results.
This method is particularly relevant in matrices where microorganisms may have been killed by heat treatment, cleaning, or disinfection but where DNA remains. Integrating Reagent D into the workflow ensures that the analysis results better reflect the actual microbiological risk, which is critical for quality control and the verification of hygiene processes.
Reagent D is used prior to DNA extraction and is compatible with foodproof® extraction systems such as the StarPrep Two Kit and the Magnetic Preparation Kit I. Following treatment, standard extraction and analysis via PCR LAMP are performed, without the need for any changes to the analytical step.
